Today I’m gonna share with you guys, the Glucose-6-phosphate dehydrogenase (G6PD) screening test (Quantitative) by the OSMMR-D-D assay, which is a new machine currently under evaluation by the Immunochemistry lab. (which is the lab I’m permanently attached to.)
WHATTT?!
So basically the G6PD is a cytoplasmic enzyme that is found dispersed in all our cells. It is involved in the hexose monophoste pathway in which it catalyzes the first step, generating NADPH which is a coenzyme that is necessary as a hydrogen donor for many biochemical pathways and for the stability of catalase, preservation and regeneration of the reduced form of glutathione. This catalase and glutathione are both necessary to detoxify hydrogen peroxide which is toxic to the cells. Thus, in order to protect the cells from hydrogen peroxide, cells are indirectly dependent on G6PD. Gluthathione helps to convert H2O2 to H2O by this enzyme known as glutathione peroxidase.
WHY!?
The clinical significance of patient with G6PD deficiency are drug-induced hemolysis, infection induced hemolysis, favism, neonatal jaundice, etc.
HOW?!!
This assay involves the use of glucose-6-phosphate dehydrogenase which will catalyze the the oxidation of glucose-6-phosphate to 6-phosphogluconate in the presence of NADP. NADPH generated then reacts with the colour reagent, reducing the tetrazolium salt within, producing a distinct colour which will be measured colourimetrically at 550nm. The colour activity has a direct relantionship with the amount of G6PD present in blood.

ReplyDeletei'm fine! thanks! how about you? anyways, the specimen used is actually blood in EDTA. :D hmmm, the pre-analytical variation are more or less the same as what we learn in LMQA. =) just that here, we have to invert the tube for a few times before pouring it into the small tube to prevent the probe from sucking up the plasma whcih will result in inaccurate results.

Anyway, if i'm not wrong, our lab sends out G6PD-deficient blood samples to your lab for quantitative measurement.
My question is: do you do qualitative or quantitative measurement to screen for presence of G6PD? I believe that the quantitative should be more expensive right?

anywaes back to your question, our quantitative measurement is still currently under evaluation.. tat means its not in used yet. In our lab, we just do qualitative measurement and if there's any deficient cases we will send to another lab (not sure if i can mention here).
i think u might have mistaken me for being in another lab. hahaha =P
anyways. its 9 more weeks! not 12! hahahha!
